چکیده

Most mutagens and genotoxic carcinogens are efficient inducers of chromosomal alterations in exposed cells. Two important classes of aberrations, namely structural and numerical, are recognized and both types of aberrations are associated with congenital abnormalities and neoplasia in humans. These alterations can be easily detected and quantified in human peripheral blood lymphocytes. Conventional staining techniques can be used to detect these aberrations; this technique was used to estimate absorbed dose in the case of a radiation accident in Goiania, Brazil. A recently introduced fluorescent in situ hybridization technique (FISH) using DNA probes has increased the sensitivity and ease of detecting chromosome aberrations, especially stable chromosome aberrations. This technique allows, to some extent, the estimation of absorbed radiation dose from past exposures. Numerical aberrations can be directly estimated in metaphases by counting the number of FISH-painted chromosomes. Micronuclei are formed by lagging chromosome fragments or whole chromosomes during the anaphase stage of cell division. The nature of micronuclei as to whether they possess a centromere can be determined either by CREST staining (calcinosis, Raynoud's phenomenon, esophageal dysmotility, sclerodactyly, telangiectasia) or FISH with centromere-specific DNA probes. In several carcinogen-exposed populations, such as heavy smokers or people exposed to arsenic, aneuploidy appears to be more common than structural aberrations. In victims of radiation accidents, aneuploidy (hyperploidy) has been found to be common in addition to structural aberrations.

Future research directions to characterize environmental mutagens in highly polluted area.

Population monitoring using methods of molecular epidemiology combined with reliable data on exposure is an extremely powerful approach to determine the effect of mutagens on human populations. Although human blood and urine have traditionally been used for biomonitoring, an increase in the use of placental and buccal smear samples should be expected. As biomarkers of exposure, DNA strand break...

Role of Cytogenetic Surveillance to Assess Exposure to Carcinogens

The hazards associated with exposure to carcinogenic and mutagenic chemicals are a growing source of public concern as well as being of research interest. Some two decades of active investigation have led to identification of carcinogens and mutagens and their sources in the human environment. Early detection of hazardous exposures may bring about significant reductions in the risk of adverse h...

Risk of cancer in an occupationally exposed cohort with increased level of chromosomal aberrations.

We used cytogenetic analysis to carry out a cohort study in which the major objective was to test the association between frequency of chromosomal aberrations and subsequent risk of cancer. In spite of the extensive use of the cytogenetic analysis of human peripheral blood lymphocytes in biomonitoring of exposure to various mutagens and carcinogens on an ecologic level, the long-term effects of...

Molecular epidemiology studies on occupational and environmental exposure to mutagens and carcinogens, 1997-1999.

Molecular epidemiology is a new and evolving area of research, combining laboratory measurement of internal dose, biologically effective dose, biologic effects, and influence of individual susceptibility with epidemiologic methodologies. Biomarkers evaluated were selected according to basic scheme: biomarkers of exposure--metabolites in urine, DNA adducts, protein adducts, and Comet assay param...

Improvement in the sensitivity of DNA polymerase I-deficient Escherichia coli for detecting mutagens and carcinogens.

The sensitivity of a polA strain to the antibacterial activity of mutagens and carcinogens may be increased by inserting one or both of the following characteristics, a lexA mutation or the R391 bacterial plasmid. The effects of the lexA mutation and the plasmid appear to be additive. The differential sensitivity of a polA lexA (R391) strain could be adapted as a preliminary screening test for ...
